I'd wait for the plants to be growing strong. Roundup washes off easy and if the plant isn't growing it won't work. At least that is how I understand it. I'd mow/burn it or clear it as best possible and then till it right now.

Okay, maybe Roundup is the wrong thing. I am thinking of something that will kill off what is presently there.

The active ingredient in Round-up is glyphosate. Generic glyphosate can work, but I've had issues with some brands in the past- specifically a Lesco branded glyphosate. Normal full-strength RoundUp is 42% active ingredient. If you have no-till seed and have a way to drill it in, I'd spray first and then plant without tilling. By tilling you are opening up more seeds to the possibility of germination as throbak mentions. Duff, as long as RoundUp has time to dry, it won't wash off with a rain. Usually it is rain-fast in around 1/2 hour, maybe a bit more if the humidity is up and the temperature is down. Safest bet is to spray on a warm sunny day! With Monsanto-branded RoundUp, you do not need any additional surfactants such as dish soap. Some of the generic ones may require additional surfactants for effective treatment- which I believe to the be the issue with the Lesco brand I used several years ago. "As always, read and follow the label directions" The label is the law, so make sure you are following it! Has the ground been tilled at all in the last couple of years or is this a completely new tilled site? If it's completely new, I'd probably spray, wait until dieback occurs (should be within 2 weeks), till and wait for any new weeds to emerge, spray, then plant within a couple of days of spraying the second time. RoundUp does not stay in active form in the soil, once it contacts soil it binds and essentially becomes inert so there is no harm in planting so soon after spraying.

Put some dawn dish soap in there too. You dont need much, but it really helps adhere the Roundup to the plant

I stand corrected! I always thought the plant needed to be growing, but with this warm weather already I don't think it would be a problem.

I use the generic glyphosate and add a little dawn. Suggest mixing the glyphosate & water first "THEN" add and mix the dawn in. Soap sud's will be a probably if you mix it in first. Yep, been there. LOL!

You are correct, the leaves obviously need to be green, and at a minimum in a slightly active growing stage. Early or late in the season I usually mix stronger than I will during the middle part of the season.

Ok, for those of you saying to "add a little dish soap".... Look at section 5 on this label, which is on page 2 on the top right hand side. Read the first paragraph!! (I'd quote it but Adobe won't let me copy and paste....): www.cdms.net/LDat/ld07A016.pdf

You don't need to spray if your breaking ground. You will turn under anything growing now. Plant roundup ready beans, spray to kill weeds, the overseed clover in the fall.

It is an old railroad bed. As far as I know it has never been tilled. It has a few weeds here and there, but it mostly leaf litter.
I am meeting the district wildlife biologist tomorrow to give me an idea of my best game plan.
