Got the bow set up.with a new drop away and new arrows. Which way does the off colored vane go, up or down?

Does it matter with a drop away rest? The rest should be out of the way before the vanes move past it.
I think it depends on how fussy you want to be with tuning your arrows, as opposed to your bow. I never really worried about that though.

With standard 100-125 gr broadheads it usually doesnt matter. With single bevels or if you just want to get really detailed you can nock tune your arrows to best performance.

I use a QAD drop away rest and I was always told the odd vane goes up, The other two would will be slight to the lest and right. This is how I have always shot. In other words the cock feather goes up.

I actually cock mine about 20 degrees from vertical. It always looks like I'm going to have clearance issues with the rest.
